小鼠犧牲登記表(Mouse Sacrifice Form)

一、操作流程：

(1)將鼠籠置於自動化犧牲箱內，基於動物福祉，建議就其原來鼠籠置入，不可混雜不同品系小鼠於同籠，亦不可彼此堆疊。

(2)先將CO₂（二氧化碳）鋼瓶頂端氣閥扭開，氣體旋即進入自動化犧牲箱。

(3)再轉開自動化犧牲箱前方啟動按鈕，綠燈亮顯示正在作用。

(4)等待10分鐘後，綠燈熄滅紅燈亮，顯示機器已作用完畢。

(5)先於外觀觀察小鼠是否完全死亡，有無微弱之呼吸或心跳。若仍有，則重複以上(2)-(4)流程。

(6)確認小鼠死亡後，先關閉CO₂（二氧化碳）鋼瓶頂端氣閥，並將自動化犧牲箱前方按鈕轉至停止位置。

(7)取出鼠籠，穿戴手套並將屍體置於本設施提供之玫瑰紙袋內（小鼠數量以玫瑰紙袋可承受重量且開口可一定程度折合為基準）。玫瑰紙袋再度裝於塑膠夾鏈袋內，丟於-20℃冰箱內之醫療廢棄物垃圾袋內。

(8)所有使用人員（P.I.、研究人員、飼育員）皆須使用本設施官網上方 > 線上申請 > 填寫「小鼠犧牲登記表」。

二、其它注意事項：

(1)由於仔鼠會閉氣，不建議使用自動化犧牲箱之CO₂犧牲方法（可能致死時間會長達30分鐘）。

(2)務必施予第二種人道處理以確保動物死亡，如：斷頸法。

1. CO₂ chamber running procedure:

(1) Place the cages in the automated CO₂ chamber. It is recommended to place mice in their original cages based on animal welfare. Mice of different strains must not be put into the same cage, nor can they be stacked on top of each other.

(2) First, open the CO₂ gas valve at the top of the cylinder, and the gas will flow in the CO₂. chamber immediately.

(3) Turn on the start button on the front of the CO₂. chamber, and the green light shows that it is on work.

(4) After 10 minutes, green light goes out and red light apperas, indicating that CO₂. chamber has completed its function.

(5) Observe the appearance of mice to see whether it is completely dead (e.g. weak breathing or heartbeat). If there are any, repeat the above (2)-(4) procedure.

(6) After confirming the mice is dead, close the CO₂ gas valve at the top of the cylinder first and turn off the button on the front of the CO₂. chamber.

(7) Take out the cages, wear gloves and place the carcass into the paper bag provided by AS core (the quantity of mice is based on the paper bag that can bear the weight and it’s opening can be folded to a certain extent). Put the paper bag into a plastic zipper bag again, and throw it into a medical waste bag in the -20°C refrigerator.

(8) All users (P.I., researcher, AS staff) must go to official website of AS core > Online application > fill in the "Mouse Sacrifice Form".

2. Other informations:

(1) Since the pups will hold their breath, it is not recommended to use CO2 sacrifice method on pups (their sacrifice period may last up to 30 minutes).

(2) Second sacrifice method must be performed to ensure the completely death of the animal, such as neck dissection.
